Microorganisms are both beneficial and harmful. These microorganisms are important to humans because they play a role in the ecology of life, by decomposing wastes, both natural and man-made, such as creating nitrogen fertilizer at the root zones of certain crops. Since unknown #5 was determined to be a Gram negative rod, an oxidase test was performed.
Oxidase Test:
For the following test, two drops of oxidase reagent were applied directly on the oxidase round paper. The unknown #5 was smeared on the oxidase paper thus created an immediate result. No color change indicated a negative test.
Indole Test:
On February 09, the unknown #5 was inoculated in the Tryptone broth. It was incubated at thirty-seven Celsius degrees for twenty-four to forty-eight hours. On February 16, the follow up period, the cultures were obtained from the refrigerator. The test tube was gently tapped to redistribute the bacteria. Twenty drops of Indole reagent were added to the test tube and then gently swirled. After the follow up, the results were immediately shown.
Urea Test:
The unknown #5 was inoculated in the urea broth tube. It was incubated at thirty-seven Celsius degrees for twenty-four to forty-eight hours on February 09, 2008. On February 16, the test tubes were observed. The results were recorded in the journal notebook.
Casein Test: The final test performed was the skim milk reaction on February 09. The unknown #5 was streaked with an inoculum needle onto the skim milk agar. It was placed in the incubator at thirty-seven Celsius degrees for twenty-four to forty-eight hours. On the next lab, the skim milk plate
